Overview

**BI 765063 + ezabenlimab + BI 836880** is an investigational triple combination therapy for the treatment of advanced solid tumors. - **BI 765063** is a monoclonal antibody antagonist of signal-regulatory protein alpha (SIRPα), blocking the CD47-SIRPα "don't eat me" immune checkpoint pathway, thereby enhancing the anti-tumor activity of myeloid cells such as macrophages and dendritic cells. - **Ezabenlimab (BI 754091)** is a monoclonal antibody checkpoint inhibitor targeting programmed cell death protein 1 (PD-1), enhancing T-cell mediated anti-tumor immune responses. - **BI 836880** is a bispecific nanobody targeting both vascular endothelial growth factor (VEGF) and angiopoietin-2 (Ang-2), inhibiting tumor angiogenesis. The combination is under investigation in advanced solid tumors including certain colorectal, endometrial, and head and neck cancers. Clinical trials indicate manageable safety and initial evidence of clinical efficacy[1][2][3]. Developed primarily by Boehringer Ingelheim, with OSE Immunotherapeutics as the original developer of BI 765063.
