BI 811283 + cytarabine is a combination regimen consisting of BI 811283, an investigational small molecule inhibitor of Aurora B kinase, and cytarabine, a well-established antimetabolite chemotherapy used primarily in the treatment of acute myeloid leukemia (AML). BI 811283 is an ATP-competitive, reversible inhibitor that selectively targets Aurora B kinase, a crucial regulator of mitosis, thus leading to mitotic checkpoint override, chromosome missegregation, and ultimately either senescence or apoptosis in proliferating tumor cells. The combination has been evaluated as a therapy for newly diagnosed AML, with BI 811283 administered as a continuous 24-hour intravenous infusion, in conjunction with subcutaneous cytarabine. Preclinical and early clinical data suggest that BI 811283 can sensitize leukemic cells to the antineoplastic effects of cytarabine, offering a rational double-hit approach to disrupt the cell cycle and DNA synthesis in AML[1][5][7].
