**BiA9*2_HF** is a preclinical, symmetric 2+2 bispecific CD40 agonistic antibody developed using Strike Pharma's Adaptive Drug Affinity Conjugate platform. It contains a human IgG2 anti-CD40 antibody derived from clone A9, with two humanized anti-pTag single-chain variable fragments fused to the CH3 domains through flexible glycine-serine linkers. The molecule agonizes CD40 on antigen-presenting cells while binding pTag-bearing tumor-antigen peptides for modular delivery, uptake, antigen presentation, and expansion of antigen-specific CD8+ and CD4+ T cells. In human-CD40 transgenic mouse tumor models, peptide-loaded BiA9*2_HF produced antigen-specific antitumor activity and showed a more favorable preclinical safety profile than selicrelumab.
