bicalutamide + aromatase inhibitor is a combination therapy being investigated for the treatment of hormone receptor-positive metastatic breast cancer. This regimen combines bicalutamide, a non-steroidal androgen receptor (AR) antagonist, with an aromatase inhibitor (AI) such as letrozole, anastrozole, or exemestane. The combination is specifically targeted at patients with estrogen receptor-positive (ER+), androgen receptor-positive (AR+), and HER2-negative disease who have experienced progression on prior AI therapy. The therapeutic rationale is based on the observation that AR signaling can mediate resistance to estrogen-targeted therapies; by simultaneously blocking both the estrogen and androgen pathways, the combination aims to restore endocrine sensitivity and improve clinical outcomes. This specific protocol is being evaluated in a Phase II clinical trial led by investigator Xu Fei at Fudan University.
