**BiCo-3 + CC-3** is a combination immunotherapy consisting of BiCo-3, a target-cell restricted bispecific anti-CD28 costimulatory antibody (BiCo), and CC-3, a B7H3xCD3 bispecific T-cell engager antibody. This pairing enhances T-cell activation, proliferation, differentiation into memory subsets, cytokine release, and potent target-restricted tumor cell lysis by recruiting CD28 costimulation alongside submitogenic CD3 engagement, specifically on tumor cells expressing B7-H3 (CD276). Developed using BiconY technology, it targets solid tumors with high B7-H3 expression, such as sarcomas and colorectal cancer, to overcome limitations of standalone T-cell engagers by improving efficacy while minimizing off-target effects; CC-3 is in Phase 1 for metastatic colorectal cancer (NCT05999396), with sarcoma trials in preparation.[1][2][4]
