Overview

Binimetinib + ganitumab is an investigational combination therapy pairing the small‑molecule MEK1/2 inhibitor binimetinib with the monoclonal antibody ganitumab, which targets the insulin‑like growth factor 1 receptor. The rationale is to concurrently inhibit MAPK pathway signaling downstream via MEK and upstream growth signaling via IGF‑1R to overcome resistance and enhance antitumor activity in advanced solid tumors. A phase 1b/2 trial (NCT01562899) was designed to establish a safe and tolerated dose of binimetinib in combination with ganitumab in selected advanced solid tumors; the study was later reported as terminated for nonmedical reasons. Binimetinib is marketed separately for other indications, whereas ganitumab has been developed as an anti‑IGF-1R antibody in oncology trials.[5][6]
