Overview

Binimetinib + nivolumab is an investigational combination therapy consisting of binimetinib, a selective small molecule inhibitor of MEK1 and MEK2 (mitogen-activated protein kinase kinases 1 and 2), and nivolumab, a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor. Binimetinib inhibits the MAPK/ERK pathway, which is often activated in cancers with RAS or BRAF mutations, thereby reducing tumor cell proliferation. Nivolumab blocks PD-1 on T cells to enhance immune-mediated antitumor responses by preventing tumor-induced immune suppression. This combination has been studied primarily in patients with metastatic colorectal cancer harboring RAS mutations and microsatellite-stable status, as well as in melanoma settings. Clinical trials have not demonstrated significant clinical benefit for this combination in RAS-mutated MSS metastatic colorectal cancer[1][6][7].
