Overview

Binimetinib + paclitaxel is a **combination therapy** of an oral MEK inhibitor, binimetinib, and an intravenous microtubule stabilizer, paclitaxel, investigated primarily for platinum-resistant epithelial ovarian cancer. Binimetinib is a highly selective, non-ATP competitive inhibitor of MEK1 and MEK2 within the MAPK pathway, while paclitaxel is a mitotic inhibitor that promotes tubulin polymerization and inhibits cell division. The combination aims for synergistic antitumor activity—MEK inhibition may sensitize tumors to taxane-induced apoptosis. The regimen’s efficacy and safety has been evaluated in phase Ib clinical trials, with the recommended phase 2 dosing of binimetinib at 30 mg orally twice daily (continuous) or 45 mg orally twice daily (intermittent), together with paclitaxel 80 mg/m^2^ intravenously weekly.
