Overview

**Binimetinib + talazoparib** is an investigational oral combination regimen studied for the treatment of metastatic pancreatic ductal adenocarcinoma (mPDAC) and potentially other advanced solid tumors. **Binimetinib** is a selective, small molecule inhibitor of MEK1/2, enzymes in the MAPK/ERK pathway, which is frequently activated in malignancies. **Talazoparib** is a small molecule poly(ADP-ribose) polymerase (PARP) inhibitor, involved in preventing the repair of single-strand DNA breaks, thereby inducing synthetic lethality in cancer cells with impaired homologous recombination repair. This combination was expected to provide additive or synergistic antitumor activity by concurrently inhibiting cell growth and proliferation pathways and enhancing DNA damage in tumor cells. Early phase Ib studies found dose-limiting toxicities at higher dosages and limited clinical efficacy in this population. The combination remains experimental and is not approved for routine clinical use[1][2][3][4].
