Overview

A combination regimen composed of **bintrafusp alfa**, **carboplatin**, and **paclitaxel** used in clinical trials for various advanced cancers. Bintrafusp alfa is a bifunctional fusion protein that simultaneously inhibits the **programmed death ligand 1 (PD-L1)** checkpoint and acts as a **trap** for the cytokine **transforming growth factor-beta (TGF-β)**. It is developed to enhance antitumor responses in the tumor microenvironment by restoring T-cell and NK-cell activity and reducing immunosuppression. **Carboplatin** and **paclitaxel** are standard cytotoxic chemotherapies, with carboplatin functioning as a DNA-damaging platinum compound and paclitaxel acting as a microtubule stabilizer to inhibit cell division. This combination has been evaluated in advanced malignancies including HPV-associated cancers, lung cancer, and biliary tract cancers, often as part of phase 1/2 clinical studies[1][3][4][5].
