Bintrafusp alfa + cisplatin + pemetrexed is a pharmaceutical combination therapy comprising **bintrafusp alfa**, a first-in-class bifunctional fusion protein targeting both transforming growth factor-beta (TGF-β) and programmed death-ligand 1 (PD-L1); **cisplatin**, a platinum-based chemotherapeutic agent; and **pemetrexed**, an antifolate chemotherapy agent. Bintrafusp alfa works by simultaneously blocking PD-L1-mediated immune evasion and sequestering TGF-β in the tumor microenvironment, which may enhance antitumor immunity and inhibit tumor-promoting effects of TGF-β[1][2][3]. Cisplatin induces DNA cross-linking leading to apoptosis, while pemetrexed inhibits folate-dependent enzymes critical for DNA synthesis. This combination is used mainly in clinical trials for advanced non-small cell lung cancer (NSCLC) and other solid tumors[1][3]. The regimen is administered intravenously.
