Biopolymer-Undisclosed (J&J) is a preclinical-stage therapeutic program being developed by Shasqi in collaboration with Johnson & Johnson. The program utilizes Shasqi's proprietary Click-to-Release platform, a two-step drug delivery system designed to localize therapeutic activity at the tumor site. The process involves the local injection of a tetrazine-modified biopolymer into the tumor, followed by the systemic administration of a protodrug—a therapeutic agent inactivated by a trans-cyclooctene (TCO) moiety. When the protodrug encounters the biopolymer, a bioorthogonal click chemistry reaction occurs, triggering the immediate release of the active payload. This localized activation is intended to increase the concentration of the drug within the tumor while reducing systemic exposure and associated toxicities. The specific payload and biological target for this collaboration remain undisclosed.
