This drug is a combination therapy for relapsed or refractory multiple myeloma (RRMM) and plasma cell leukemia, developed by the University of Würzburg. It consists of a biparatopic nanobody-based B cell maturation antigen (BCMA) chimeric antigen receptor T (CAR-T) cell therapy and the immunomodulatory drug pomalidomide. The CAR-T component is designed to simultaneously engage two non-overlapping BCMA epitopes, demonstrating superior cytotoxicity and sustained killing compared to existing BCMA CAR-T therapies in preclinical studies. Pomalidomide, a small molecule, augments T cell function, promotes memory-like differentiation, and prolongs effector persistence, synergizing with the CAR-T cells. Pomalidomide's mechanism involves binding to Cereblon, leading to the degradation of Ikaros family zinc finger protein 1 and 3.
