BL-M07D1 + pertuzumab + docetaxel is an investigational combination regimen consisting of BL-M07D1 (trastuzumab brengitecan), pertuzumab, and docetaxel. BL-M07D1 is an antibody-drug conjugate targeting HER2, comprising a trastuzumab-derived monoclonal antibody, a cathepsin B-cleavable linker, and the topoisomerase I inhibitor payload Ed-04 (a camptothecin derivative). Pertuzumab is a humanized monoclonal antibody that blocks HER2 dimerization, while docetaxel is a cytotoxic taxane chemotherapy. This triple regimen is under clinical investigation primarily for HER2-positive breast cancer and is being evaluated for safety and efficacy as first-line therapy in the metastatic or unresectable locally advanced setting. BL-M07D1 demonstrates strong preclinical tumor suppression, including against low HER2-expressing tumors, mediated by inhibition of topoisomerase I and induction of cell cycle arrest and apoptosis. Preliminary clinical data show tolerability of the combination, with adverse events mainly related to myelosuppression and mucosal toxicity typical for the class[1][3][5][7][9].
