Bld-1-D(KLAKLAK)2 is an experimental peptide-drug conjugate (PDC) designed for the targeted treatment of bladder cancer. It comprises a bladder tumor-targeting peptide, Bld-1 (sequence: CSNRDARRC), conjugated to a pro-apoptotic D-enantiomer peptide, D(KLAKLAK)2. The Bld-1 moiety serves as a homing domain that selectively binds to bladder tumor cells and facilitates cellular internalization. Once inside the cell, the D(KLAKLAK)2 peptide induces apoptosis by disrupting mitochondrial membranes. Preclinical research has shown that Bld-1-D(KLAKLAK)2 exerts dose- and time-dependent cytotoxic effects on HT-1376 bladder tumor cells and significantly inhibits tumor growth in mouse models, demonstrating superior efficacy compared to the pro-apoptotic peptide alone.
