Overview

This combination therapy is utilized during the re-induction phase of the SWOG S2106 clinical trial for patients with newly diagnosed Philadelphia chromosome-positive (Ph+) acute lymphoblastic leukemia (ALL). The regimen consists of **blinatumomab**, a bispecific T-cell engager (BiTE) that simultaneously targets CD19 on B-cells and CD3 on T-cells to facilitate immune-mediated tumor cell lysis; **dasatinib**, a second-generation tyrosine kinase inhibitor (TKI) that targets the BCR-ABL1 fusion protein characteristic of Ph+ ALL; **methotrexate**, an antimetabolite that inhibits dihydrofolate reductase (DHFR) to disrupt DNA synthesis; and **dexamethasone**, a potent corticosteroid with lympholytic and anti-inflammatory properties. This multi-targeted approach is designed to achieve deep molecular remission by combining targeted molecular therapy, immunotherapy, and traditional chemotherapy.
