Overview

Blinatumomab + venetoclax is an investigational combination therapy for hematologic malignancies, primarily relapsed or refractory B-cell precursor acute lymphoblastic leukemia (BCP-ALL) and minimal residual disease (MRD)-positive BCP-ALL. Blinatumomab is a bispecific T-cell engager antibody construct that targets CD19 on B cells and CD3 on T cells, redirecting cytotoxic T cells to eliminate malignant B cells. Venetoclax is a small molecule inhibitor of the anti-apoptotic protein BCL-2, promoting apoptosis in cancerous lymphoid cells. The combination aims to enhance anti-leukemic efficacy by leveraging both immune-mediated cytotoxicity and direct induction of apoptosis. Early-phase clinical trials have shown promising activity in patients with high leukemia burden or poor prognosis who are not responsive to standard therapies[1][2][8].
