Overview

BMS-663068 + ritonavir is an investigational combination therapy for HIV-1 infection, involving **BMS-663068**, a prodrug of the small molecule inhibitor BMS-626529, and **ritonavir**. BMS-626529 acts as a first-in-class HIV-1 attachment inhibitor by binding the viral envelope protein gp120, preventing initial attachment of HIV to host CD4+ T cells and subsequent viral entry. Ritonavir is included as a pharmacokinetic enhancer (booster) due to its potent inhibition of CYP3A4, which increases plasma concentrations of BMS-626529, improving antiviral activity. This combination has demonstrated substantial reductions in HIV-1 RNA, good safety/tolerability, and is being tested primarily in highly treatment-experienced individuals with multidrug-resistant HIV-1[1][2][3][7][8].
