Overview

**BMS-754807 + metformin** is a combination of two small molecules: BMS-754807, a potent, reversible inhibitor of the insulin-like growth factor 1 receptor (IGF-1R) and insulin receptor (IR), and metformin, a widely used biguanide antidiabetic agent that also exhibits pleiotropic anti-tumor activities via modulation of metabolic and cell signaling pathways. Studies in triple-negative breast cancer (TNBC) cell lines demonstrate that the combination shows synergistic antiproliferative effects in most tested models, mechanistically associated with increased levels of the cell cycle inhibitor p27^Kip1^ (CDKN1B) and decreased levels of the E3 ubiquitin ligase SCF^Skp2^. The combination's enhanced efficacy versus either agent alone suggests that it targets cancer growth through both metabolic and growth factor signaling pathways[1][3].
