Overview

BMS-754807 + trastuzumab is an investigational combination therapy composed of BMS-754807, a small molecule, orally available, reversible dual inhibitor of insulin-like growth factor 1 receptor (IGF-1R) and insulin receptor (IR) family kinases, and trastuzumab (marketed as Herceptin), a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2/ERBB2). BMS-754807 blocks IGF-1R and IR kinase signaling, interfering with growth and survival pathways in cancer cells, while trastuzumab binds to HER2 and inhibits its signaling, inducing cellular cytotoxicity. This combination is being evaluated primarily for advanced or metastatic HER2-positive breast cancer, especially in cases resistant to trastuzumab monotherapy, with the goal of overcoming resistance through dual pathway inhibition[1][3][5].
