Overview

**BMS-906024 + carboplatin + paclitaxel** is an experimental combination regimen evaluated primarily in preclinical and early clinical studies for the treatment of certain solid tumors, including non-small cell lung cancer (NSCLC). BMS-906024 is a potent, selective, orally bioavailable gamma secretase inhibitor that blocks Notch signaling by inhibiting gamma secretase activity required for activation of all four Notch receptors. Notch pathway inhibition has been implicated in overcoming resistance and reducing proliferation in cancer cells. Preclinical studies show that combining BMS-906024 with paclitaxel and/or carboplatin enhances anti-tumor effects, particularly in KRAS- and BRAF-wild type NSCLC models, through increased apoptosis and decreased cell proliferation. Addition of BMS-906024 to paclitaxel or carboplatin leads to synergistic cytotoxicity compared to chemotherapy alone. BMS-906024 was developed by Bristol Myers Squibb, and the combination is still considered investigational and not established for clinical use[1][2][4].
