BMS-906024 + paclitaxel is a combination regimen consisting of BMS-906024—a potent, selective pan-Notch gamma secretase inhibitor—and paclitaxel, a widely used cytotoxic microtubule stabilizing chemotherapy. BMS-906024 inhibits activation of all four Notch receptors, functioning by blocking the presenilin-dependent gamma secretase complex, thus preventing Notch receptor cleavage and pathway activation. The combination has been shown to synergistically enhance antitumor activity in preclinical models of non-small cell lung cancer (NSCLC), particularly in tumors with wild-type KRAS and BRAF, leading to greater cell apoptosis and reduced proliferation compared to either agent alone. BMS-906024 is primarily under investigation for sensitizing solid tumor cells and leukemia to chemotherapy, with a focus on lung adenocarcinoma[1][3].
