Overview

**BMS-908662 + ipilimumab** is a combination therapy of a pan-RAF inhibitor (BMS-908662) and a CTLA-4 blocking monoclonal antibody (ipilimumab). BMS-908662 targets multiple RAF isoforms (B-RAF^V600^, B-RAF^wt^, A-RAF, and C-RAF), disrupting the MAPK signaling pathway involved in cancer cell proliferation, and is administered orally. Ipilimumab is an immune checkpoint inhibitor that blocks CTLA-4, enhancing T-cell activation and antitumor immune response; it is administered intravenously. This combination was studied in phase 1 trials for unresectable stage III or IV melanoma, particularly melanoma with BRAF^V600^ mutations, to evaluate safety and additive or synergistic antitumor activity by combining targeted therapy with immunotherapy[1][3][5][7].
