Overview

BMS-986183 + nivolumab is a combination investigational therapy consisting of BMS-986183, an antibody-drug conjugate (ADC) targeting Glypican-3 (GPC3), and nivolumab, a programmed death-1 (PD-1) immune checkpoint inhibitor. BMS-986183 acts by selectively binding to GPC3-expressing tumor cells (primarily seen in hepatocellular carcinoma) and delivering a cytotoxic payload (tubulysin) via its ADC modality, leading to targeted cell death. Nivolumab enhances anti-tumor immunity by blocking the PD-1 receptor on T-cells, preventing tumor-induced T-cell inhibition. This combination was developed for advanced hepatocellular carcinoma not amenable to curative treatment, investigating overall safety, tolerability, anti-tumor activity, and pharmacokinetics[1][2][3].
