**BMS-986256 + famotidine** is a combination of afimetoran (BMS-986256), an orally bioavailable, first-in-class, selective small molecule inhibitor of Toll-like receptor 7 (TLR7) and Toll-like receptor 8 (TLR8), and famotidine, a small molecule histamine H2 receptor antagonist that inhibits gastric acid secretion. Afimetoran is being developed primarily for systemic lupus erythematosus (SLE) and cutaneous lupus erythematosus (CLE); famotidine is widely used in gastrointestinal indications such as peptic ulcers and gastroesophageal reflux disease (GERD). The combination has been evaluated in clinical pharmacology studies to determine whether famotidine affects the pharmacokinetics of afimetoran, as famotidine can alter gastric pH and drug absorption. There is no indication this combination is an approved product or is under development as a fixed-combination therapy; rather, it has been co-administered in early-phase studies for interaction assessment[3][9].
