Overview

BMS-986299 + nivolumab + ipilimumab is an experimental triple combination regimen for advanced solid tumors. **BMS-986299** is a first-in-class, intratumorally delivered NLRP3 inflammasome agonist that enhances adaptive immune and T-cell memory responses by activating the NLRP3 inflammasome, which leads to increased release of pro-inflammatory cytokines such as IL-1β, G-CSF, and IL-6 and stimulates tumor-infiltrating lymphocytes. **Nivolumab** is a monoclonal antibody targeting Programmed cell death protein 1 (PD-1), functioning as a checkpoint inhibitor to restore T-cell activity in the tumor microenvironment. **Ipilimumab** is a monoclonal antibody against Cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), also relieving inhibitory signals on T cells. The triple combination is intended to synergistically modulate the tumor immune microenvironment, enhancing antitumor immunity particularly in patients who have previously failed or are refractory to standard immunotherapies. Early-phase clinical studies (phase I) demonstrated manageable safety and promising activity in some cancer types, notably advanced and immunotherapy-refractory breast, squamous cell, and head and neck cancers[1][2][3][4][5][6].
