Overview

BMS-986360 + nivolumab is a combination drug being investigated primarily for the treatment of advanced solid tumors, including triple-negative breast cancer, lung cancer, colorectal cancer, head and neck cancer, renal cell carcinoma, pancreatic cancer, sarcoma, and others. BMS-986360 is a small molecule, oral, reversible pan–c-Jun N-terminal kinase (JNK) inhibitor that targets JNK signaling pathways to modulate the tumor microenvironment and potentiate antitumor immune responses. Nivolumab is a monoclonal antibody checkpoint inhibitor targeting programmed cell death protein 1 (PD-1), blocking the immune checkpoint and thus enhancing cytotoxic T-cell responses against tumor cells. The rationale for this combination is synergistic activation of the immune system: inhibition of JNK may increase tumor immunogenicity and foster responsiveness to immunotherapy, while nivolumab controls the immune checkpoint and amplifies immune-mediated tumor cell death. The combination is under clinical investigation, primarily by Bristol Myers Squibb, in phase 1 trials for advanced cancers, with safety, tolerability, and preliminary efficacy being the main outcomes measured[1][3][5][6][7].
