BMS-986393 + mezigdomide is an investigational drug combination being studied in relapsed and/or refractory multiple myeloma. BMS-986393 (arlocabtagene autoleucel, also known as CC-95266) is an autologous chimeric antigen receptor (CAR) T cell therapy targeting GPRC5D, a protein expressed on malignant plasma cells. Mezigdomide (CC-92480) is a novel cereblon E3 ligase modulator (CELMoD) and protein degrader; it acts by stimulating the immune system and targeting degradation of tumor-promoting proteins, promoting direct cytotoxicity to multiple myeloma cells. This combination aims to improve anti-myeloma activity by simultaneously employing cell therapy and protein degradation mechanisms. Both drugs are being developed and studied in combination for patients with multiple myeloma who have failed previous standard therapies[1][3][4][5][9].
