Overview

**BMS-986406 + nivolumab** is an investigational combination therapy consisting of BMS-986406, a monoclonal antibody targeting leukocyte immunoglobulin-like receptor subfamily B member 2 (LILRB2 or ILT-4), and nivolumab, a programmed death-1 (PD-1) immune checkpoint inhibitor. BMS-986406 works by inhibiting LILRB2, a receptor involved in suppressing anti-tumor immune responses, thereby potentially enhancing immune-mediated tumor killing. Nivolumab blocks the interaction between PD-1 on immune cells and its ligands on tumor cells, which prevents downregulation of immune activity, further promoting anti-tumor immunity. This combination has been studied for safety and tolerability in patients with advanced solid tumors, including metastatic renal cell carcinoma, castration-resistant prostate cancer, ovarian cancer, melanoma, non-small cell lung cancer, pancreatic ductal adenocarcinoma, gastric cancer, squamous cell carcinoma of the head and neck, gastroesophageal junction cancer, bladder cancer, mesothelioma, triple-negative breast cancer, and soft tissue sarcoma[1][2][4][6].
