Overview

BMS-986408 + nivolumab + ipilimumab is an investigational **combination therapy** being evaluated in advanced solid tumors. **BMS-986408** is a first-in-class **dual diacylglycerol kinase alpha and zeta (DGKα/ζ) inhibitor**, developed to enhance T cell signaling and antitumor immunity by blocking negative regulation through diacylglycerol metabolism. **Nivolumab** is a monoclonal antibody targeting **programmed cell death protein 1 (PD-1)**, functioning as a checkpoint inhibitor to enhance immune responses against tumors. **Ipilimumab** is a monoclonal antibody targeting **cytotoxic T-lymphocyte–associated antigen 4 (CTLA-4)**, another checkpoint inhibitor, designed to further stimulate immune activation. The rationale of this combination is that DGKα/ζ inhibition may potentiate the therapeutic effects of PD-1 and CTLA-4 blockade, leading to improved T-cell–mediated antitumor responses. The combination is under evaluation for safety, tolerability, and optimal dosing in patients with advanced solid tumors, including those refractory to standard therapies[1][2][3][5][6][8].
