Overview

BMS-986416 + nivolumab is an investigational combination therapy comprising BMS-986416, a TGF-beta receptor-based TGF-beta trap that neutralizes TGF-beta1 and TGF-beta3, and nivolumab (Opdivo), a monoclonal antibody targeting PD-1 (programmed cell death protein 1). BMS-986416 acts by blocking TGF-beta1 and TGF-beta3 without affecting TGF-beta2, leading to decreased downstream signaling and potentially enhanced anti-tumor T cell responses. Nivolumab inhibits PD-1, resulting in increased T-cell activation and enhanced anti-tumor immune activity. This combination is under clinical investigation (Phase 1) for use in individuals with advanced solid tumors resistant or intolerant to standard therapies, including non-small cell lung cancer, urothelial carcinoma, squamous cell carcinoma of the head and neck, hepatocellular carcinoma, microsatellite-stable colorectal carcinoma, and pancreatic ductal adenocarcinoma[1][3][5].
