BMS-986442 + nivolumab + docetaxel is an experimental combination therapy for advanced solid tumors including non-small cell lung cancer. BMS-986442 is a bispecific monoclonal antibody that targets and inhibits both TIGIT (T cell immunoglobulin and ITIM domains) and CD96 (also known as Tactile), immune checkpoint proteins implicated in tumor-mediated immune suppression. Nivolumab is a PD-1 (programmed death-1) immune checkpoint inhibitor monoclonal antibody that enhances T-cell–mediated antitumor responses. Docetaxel is a cytotoxic chemotherapy agent belonging to the taxane class, which stabilizes microtubules and inhibits cell division. The rationale for the combination is that dual immune checkpoint blockade (TIGIT/CD96 and PD-1) together with cytotoxic chemotherapy may synergistically augment antitumor immunity and efficacy, especially in cancers resistant to immune monotherapy. The combination is being investigated in Phase 1/2 trials for solid tumors and non-small cell lung cancer, but the combination with chemotherapy (including docetaxel) in these trials was not initiated before study termination due to business reasons and not safety issues[3][2][4][6][5].
