BMS-986449 + nivolumab is an experimental combination therapy for advanced solid tumors. BMS-986449 is a potent, orally bioavailable small molecule degrader targeting IKZF2 and IKZF4 proteins, with minimal impact on IKZF1/3 in regulatory T-cells, and has been developed as an immunomodulatory agent[4]. Nivolumab (brand name Opdivo) is a monoclonal antibody directed against the immune checkpoint protein PD-1, blocking its interaction with PD-L1 and PD-L2 to enhance anti-tumor immune responses. The rationale for the combination arises from targeting two separate axes of immune regulation: immunomodulation via IKZF2/4 degradation (potentially modifying the tumor microenvironment and T-cell response) and checkpoint inhibition via PD-1 blockade. Early-phase clinical trials are currently assessing safety and efficacy in advanced solid tumors, including non-small cell lung cancer (NSCLC) and triple-negative breast cancer (TNBC)[1][2][3][4][5][6][7][9].
