Drug intelligence / Profile preview
BMS-986453 is a small molecule inhibitor of the transient receptor potential canonical 4 and 5 (TRPC4/5) ion channels. Originally developed through a collaboration between Sanofi and Genentech (as SAR441255), the program was later acquired or licensed by Bristol Myers Squibb. TRPC4 and TRPC5 are non-selective cation channels expressed in brain regions associated with emotional processing, such as the amygdala and hippocampus. By inhibiting these channels, BMS-986453 aims to modulate neuronal excitability and synaptic plasticity, potentially offering a novel therapeutic approach for mood and anxiety disorders. The drug is currently in Phase 1 clinical development, where its safety, tolerability, and pharmacokinetics are being evaluated in healthy participants.
