Overview

BMX-001 + temozolomide is a combination therapy under investigation for the treatment of newly diagnosed high-grade glioma. BMX-001 is a metalloporphyrin small molecule that mimics the action of superoxide dismutase, an enzyme that neutralizes free radicals and protects cells from oxidative stress. It acts as both a radioprotectant for healthy tissue and a radiosensitizer for tumor cells, enhancing the effects of radiation therapy and chemotherapy while reducing side effects such as cognitive decline. The mechanism involves inhibition of NFkB and HIF-1a signaling pathways, which are associated with inflammation, cell survival, and angiogenesis in tumors. Temozolomide is an oral alkylating agent used as standard-of-care chemotherapy in high-grade glioma. In phase 2 clinical trials, this combination has demonstrated increased median survival by 6.6 months compared to standard chemoradiation alone, along with mitigation of radiation-induced cognitive issues and improved white matter integrity[3][4][5][8].
