BND-35 + nivolumab is an experimental combination therapy for advanced solid tumors composed of BND-35, a novel fully human monoclonal antibody that antagonizes ILT3/LILRB4 on myeloid cells to remodel the immunosuppressive tumor microenvironment, plus nivolumab, an approved anti-PD-1 immune checkpoint inhibitor. BND-35 blocks interactions of ILT3/LILRB4, thereby enhancing antibody-dependent cellular cytotoxicity and phagocytosis, reactivating T cells and NK cells suppressed by tumors, and promoting a pro-inflammatory tumor milieu. Nivolumab further potentiates anti-tumor immune response by targeting the PD-1 receptor on T cells, preventing tumor-induced T cell inhibition. The combination's rationale is the synergistic reversal of immunosuppression and augmentation of tumor cell destruction, particularly in cancers resistant to current immunotherapies. BND-35 is in early-phase (as of 2025) clinical development in combination with nivolumab for advanced solid tumors[1][2][4][5][6][8].
