BNT112 + cemiplimab is a **combination therapy** composed of BNT112, a liposome-complexed messenger RNA (mRNA) cancer vaccine targeting five specific prostate cancer antigens, and cemiplimab, a monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1) immune checkpoint. BNT112 is designed to induce antigen-specific CD8+/CD4+ T-cell responses via efficient antigen presentation and innate immune activation through Toll-like receptor 7 (TLR7) agonism, while cemiplimab enhances T-cell mediated anti-tumor immunity by blocking PD-1-mediated immune suppression. The combination aims for synergistic immunostimulation and immune checkpoint inhibition, and is being developed primarily for prostate cancer (both metastatic castration resistant and high-risk localized) with possible application in other tumor types.
