BNT116 + BNT316 is an investigational combination therapy being evaluated for advanced non-small cell lung cancer (NSCLC). BNT116 is an mRNA-based cancer vaccine designed to encode six fixed, tumor-associated antigens (MAGE-A3, MAGE-A4, MAGE-C1, Claudin6, KK-LC-1, and PRAME) to prime the immune system against NSCLC cells. BNT316 is an anti-cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) monoclonal antibody intended to enhance immune activation by inhibiting a key immune checkpoint, thereby promoting T-cell-mediated anti-tumor responses. The combination leverages both targeted immunization (via mRNA-encoded antigens) and checkpoint inhibition (via anti-CTLA-4), aiming to induce a robust immune attack on tumor cells. The combination is currently in early-phase clinical trials for NSCLC, primarily under development by BioNTech[1][2][3][5].
