BNT142 is an investigational messenger RNA (mRNA) vaccine developed by BioNTech that targets fibroblast activation protein alpha (FAP). FAP is a cell-surface serine protease highly expressed on cancer-associated fibroblasts (CAFs), which play a crucial role in promoting tumor growth, immunosuppression, and metastasis within the tumor microenvironment. By delivering mRNA encoding FAP, the vaccine aims to induce a robust T-cell-mediated immune response against CAFs, thereby disrupting the supportive tumor stroma and enhancing the efficacy of other immunotherapies. BNT142 is formulated in lipid nanoparticles (LNPs) for systemic administration and is currently being evaluated in Phase 1/2 clinical trials for patients with advanced solid tumors.
