Overview

BNT326 + paroxetine is a **combination of BNT326**, an investigational antibody-drug conjugate (ADC) targeting human epidermal growth factor receptor 3 (HER3), and **paroxetine**, an approved selective serotonin reuptake inhibitor (SSRI). BNT326 (also known as YL202) consists of an anti-HER3 antibody linked to a topoisomerase I inhibitor via a protease-cleavable linker, inducing DNA damage–mediated apoptosis specifically in HER3-expressing tumor cells. Paroxetine is primarily used as an antidepressant and acts by inhibiting serotonin reuptake in the central nervous system. This combination has been studied in a dedicated drug-drug interaction (DDI) cohort to assess the effect of CYP inhibition (paroxetine is a strong CYP2D6 inhibitor) on BNT326 pharmacokinetics. The combination is not a therapeutic regimen per se but is tested to characterize pharmacokinetic interactions, not for a direct anti-cancer indication[3].
