BNT327 + carboplatin + pemetrexed is an investigational combination therapy for the treatment of solid tumors, particularly being studied as a first-line therapy for extensive-stage small cell lung cancer (ES-SCLC) and non-small cell lung cancer (NSCLC). This regimen consists of BNT327, a next-generation bispecific antibody targeting both PD-L1 (Programmed Death-Ligand 1) and VEGF-A (Vascular Endothelial Growth Factor A), combined with the chemotherapy agents carboplatin and pemetrexed. BNT327 works by simultaneously blocking PD-L1, to enhance T-cell–mediated immune responses against tumor cells, and neutralizing VEGF-A, to inhibit tumor angiogenesis and modulate the immunosuppressive tumor microenvironment. The combination with carboplatin, a platinum-based DNA-damaging agent, and pemetrexed, an antifolate antineoplastic that disrupts folate-dependent metabolic processes essential for cell replication, aims to further improve anti-tumor activity. This combination has shown strong preliminary efficacy and a manageable safety profile in ongoing phase 2 and 3 clinical trials[1][2][3][4][5][6][7][8].
