Overview

BNT327 + etoposide + carboplatin is a combination therapy under late-phase clinical investigation for extensive-stage small-cell lung cancer (ES-SCLC). It comprises **BNT327**, a bispecific monoclonal antibody targeting PD-L1 and VEGF-A, alongside **etoposide** and **carboplatin**, two standard cytotoxic chemotherapeutics. BNT327 is designed to simultaneously inhibit PD-L1, restoring cytotoxic T cell activity against tumor cells, and neutralize VEGF-A, disrupting tumor angiogenesis and modulating the immunosuppressive tumor microenvironment. Etoposide inhibits topoisomerase II, causing DNA strand breaks and tumor cell death; carboplatin induces DNA cross-linking, culminating in apoptosis. The combination showed an overall response rate of 85.4% in phase 2 and is currently being evaluated in phase 3 trials as first-line therapy for ES-SCLC[2][4][7][9]. BNT327 is co-developed by BioNTech and Bristol Myers Squibb, and has a manageable safety profile with hematologic and immune-related adverse events consistent with its mechanisms[2][4][6].
