Overview

This is a combination of **bococizumab**, a humanized monoclonal antibody that targets **proprotein convertase subtilisin/kexin type 9 (PCSK9)**, and **recombinant human hyaluronidase (rHuPH20)**, an enzyme enhancing subcutaneous (SC) absorption and dispersion of co-administered therapeutics. Bococizumab inhibits PCSK9, leading to increased LDL receptor levels and thus lowering LDL-cholesterol. rHuPH20 hydrolyzes hyaluronan in the extracellular matrix, temporarily increasing SC tissue permeability and facilitating delivery of larger volumes or concentrations of injected drugs. The combination was investigated to allow less frequent dosing and improved bioavailability of bococizumab. The clinical development of bococizumab was discontinued, and the combination did not demonstrate a proportional increase in lipid-lowering efficacy, despite increased bioavailability.
