Overview

This refers to a combination therapy regimen consisting of bortezomib, cytarabine, and dexamethasone, which was evaluated in a Phase 2 clinical trial (NCT01793584) led by Seoul National University Hospital for the treatment of relapsed or refractory mantle cell lymphoma (MCL). The regimen combines the proteasome-inhibiting activity of bortezomib, which induces cell cycle arrest and apoptosis, with the antimetabolite effects of cytarabine, which inhibits DNA synthesis. Dexamethasone, a potent synthetic glucocorticoid, is included for its lympholytic properties and its ability to enhance the efficacy of the other agents. The trial specifically targeted patients who had failed 1 to 3 prior lines of therapy and utilized pegteograstim (or pegfilgrastim) for myeloid support to manage hematologic toxicity. Results from this study were published in the British Journal of Haematology in 2023.
