Bortezomib + dexamethasone is a combination therapy used primarily in the treatment of multiple myeloma, especially when other treatments have failed or the disease has relapsed. Bortezomib is a small molecule proteasome inhibitor that targets the 26S proteasome, disrupting protein degradation and leading to cell cycle arrest and apoptosis in malignant cells. Dexamethasone is a synthetic glucocorticoid corticosteroid with anti-inflammatory and immunosuppressant properties; it also induces apoptosis in certain cancer cells and enhances the efficacy of bortezomib. The combination exploits complementary mechanisms—proteasome inhibition by bortezomib and steroid-induced cytotoxicity by dexamethasone—to maximize anti-myeloma activity[5][6][1]. This regimen may be administered as part of first-line or relapsed/refractory multiple myeloma protocols.
