Bortezomib + doxorubicin is a combination therapy investigated for the treatment of metastatic breast cancer. This regimen combines **bortezomib**, a first-in-class reversible inhibitor of the 26S proteasome, with **doxorubicin**, an anthracycline antineoplastic agent. Bortezomib works by blocking the degradation of intracellular proteins, leading to the accumulation of pro-apoptotic factors and inhibition of the NF-κB pathway, which can sensitize tumor cells to the cytotoxic effects of chemotherapy. Doxorubicin exerts its effect by intercalating into DNA and inhibiting topoisomerase II, resulting in DNA double-strand breaks and cell death. The combination was evaluated in a Phase II clinical trial (NCT00101660) led by the **University of Wisconsin, Madison**, specifically targeting patients with metastatic breast cancer who had received minimal prior chemotherapy. The rationale for this combination is to exploit potential synergy between proteasome inhibition and DNA-damaging agents to overcome chemoresistance.
