PAD induction is a combination chemotherapy regimen consisting of bortezomib, doxorubicin, and dexamethasone. It is primarily used as induction therapy for patients with newly diagnosed multiple myeloma who are candidates for autologous stem cell transplant (ASCT). Bortezomib is a proteasome inhibitor that induces apoptosis by blocking the degradation of pro-apoptotic proteins. Doxorubicin is an anthracycline that intercalates into DNA and inhibits topoisomerase II, while dexamethasone is a corticosteroid that provides synergistic anti-myeloma activity and helps manage inflammation. The regimen was notably evaluated in the Phase II PIMMS trial (NCT00872521) sponsored by Janssen-Cilag Pty Ltd.
