Overview

Bortezomib + RhB-MOF is a combination nanomedicine where bortezomib, a proteasome inhibitor, is encapsulated within an iron-based metal-organic framework (MOF) modified by rhodamine B (RhB). This formulation—known as BTZ@RhB-MOF—enables targeted delivery to tumor cells, particularly colorectal cancer cells, by facilitating the release of both bortezomib and iron ions under acidic tumor microenvironment conditions. When internalized by cancer cells, BTZ@RhB-MOF localizes to mitochondrial outer membranes, binds to TOM20, destroys mitochondrial function, and induces ferroptosis through increased intracellular iron and suppression of mitochondrial protein function, resulting in anti-tumor effects in vitro and in vivo[2].
