Overview

BP1001 + dasatinib is a combination therapy being clinically investigated for the treatment of Philadelphia chromosome-positive (Ph+) chronic myelogenous leukemia (CML), Ph+ acute myeloid leukemia (AML), and high-risk Ph+ myelodysplastic syndromes (MDS)[1][3][7]. BP1001 is a liposomal Grb2 antisense oligonucleotide that inhibits the expression of Grb2, an adaptor protein important for signal transduction from oncogenic tyrosine kinases. Its inhibition is thought to disrupt survival and proliferation of leukemia cells. Dasatinib is an oral BCR-ABL tyrosine kinase inhibitor that blocks signaling from the Philadelphia chromosome fusion gene, thereby directly inhibiting leukemia cell growth[2][4][8]. The combination intends to simultaneously inhibit critical signaling pathways and enhance antileukemic efficacy. The main indication under investigation is for Ph+ hematologic malignancies, especially in patients who have failed initial tyrosine kinase inhibitor therapy[1][3][7].
