Overview

BPX-101 + AP1903 is a **combination immunotherapy** for metastatic castration-resistant prostate cancer (mCRPC), comprising an autologous dendritic cell (DC)-based vaccine (BPX-101) and the small molecule dimerizer rimiducid (AP1903). BPX-101 is manufactured by transducing autologous antigen-presenting cells with an adenoviral vector encoding an inducible CD40 (ih-CD40) protein and loading them with prostate-specific membrane antigen (PSMA) peptides. Upon intradermal administration, BPX-101 is activated in vivo by intravenous rimiducid, which cross-links the modified CD40, leading to localized, temporally controlled DC activation. The overall mechanism is to induce a potent, antigen-specific T-cell response and systemic immune activation against PSMA-expressing prostate tumor cells[1][3][5][7].
